TBS-T appears as white to off-white instant granules. Each pouch of TBS-T instant granules is sufficient to prepare 1 L of 1× TBS-T buffer, featuring simple and convenient operation.The main components of TBS-T are sodium chloride, potassium chloride, Tris buffer salts and Tween-20. The 1× TBS-T buffer contains 137 mM sodium chloride, 2.7 mM potassium chloride, 25 mM Tris buffer salts and 0.05% Tween-20. The pH value of the prepared 1× solution is 7.4±0.15 at 25℃.
As a widely used biological buffer, TBS-T is mainly applied in immunohistochemistry, in situ hybridization (ISH), enzyme-linked immunosorbent assay (ELISA) and Western blotting. It is commonly used as a washing buffer, and also frequently for antibody dilution and blocking solution preparation.
Protocol
1. Measure approximately 600 mL of distilled water into a beaker, and place a magnetic stir bar in the beaker.
2. Place the beaker on a magnetic stirrer. Slowly add the entire contents of one pouch of TBS-T instant granules, and stir until completely dissolved.
3. Add distilled water to the TBS-T solution from Step 2 to a final volume of 1 L to obtain 1× working solution.
