Acid Phosphatase (ACP) is widely distributed throughout various tissues and is primarily located in cellular lysosomes, making it a common lysosomal marker enzyme. Extralysosomal acid phosphatase exists in the endoplasmic reticulum and cytoplasm. Acid phosphatases vary among different animals, and their optimal pH range is 4.5-5.5. Acid phosphatase is a protein family, with molecular weights ranging from 18 kDa to 100 kDa in mammals. The enzyme is classified into two types: one is tartrate-sensitive, and the other is fluoride-sensitive. Acid phosphatase in lysosomes is tartrate-sensitive, while acid phosphatase in red blood cells and macrophages is fluoride-sensitive. The detection principle of this kit is as follows: Using disodium phenyl phosphate as a substrate, it is hydrolyzed by acid phosphatase (ACP) to generate phenol and inorganic phosphorus. Under alkaline conditions, phenol reacts with 4-AAP, and the product is oxidized to form a red quinone compound. The intensity of the red color is positively correlated with acid phosphatase activity; higher activity results in a deeper red color, while lower activity results in a lighter color. By measuring the absorbance at 510 nm using a microplate reader, the acid phosphatase activity in the sample can be calculated. This kit is primarily used for detecting prostatic acid phosphatase activity in serum, and is also suitable for measuring endogenous acid phosphatase activity in samples such as cell or tissue lysates or homogenates, plasma, serum, and urine. This product is for research use only and is not intended for use in diagnostic procedures.
