The cell wall of mycobacteria contains a large amount of lipids that encase the peptidoglycan layer, which is why mycobacteria are generally difficult to stain. Traditional staining methods require heating and extended staining time to facilitate dyeing. Once mycolic acid in mycobacteria binds to the dye, it is resistant to decolorization by acidic decolorizing solution — hence the name acid-fast staining. The Acid-fast Stain Solution (Modified Kinyoun Cold Staining Method) is a cold-type stain that does not require heating, making it relatively safer than heat-based acid-fast stain solutions. Its staining principle is as follows: at room temperature, mycolic acid combines with fuchsin to form a stable complex. After counterstaining with methylene blue, mycobacteria remain bright red, while other bacteria and background substances are stained blue.

Operating Procedures (For Reference Only):
1. Pick the test sample with an inoculating loop, spread it evenly on a glass slide, and air-dry naturally.
2. Add a drop of Modified Kinyoun Fuchsin Stain Solution and perform staining.
3. Heating is mandatory. Rinse the slide with distilled water.
4. Decolorize the slide with Kinyoun Decolorizing Solution.
5. Rinse with distilled water.
6. Stain the slide with Methylene Blue Stain Solution.
7. Rinse with distilled water.
8. Blot the water gently and air-dry naturally.
9. Examine the slide under an oil immersion lens.
Staining Results:
Acid-fast or weakly acid-fast bacteria: Red
Non-acid-fast bacteria, cells, and background: Light blue

1. Tighten the reagent bottle caps immediately after each use to prevent reagent volatilization and contamination.
