Carnitine palmitoyltransferase is a class of acyltransferases located in the inner mitochondrial membrane. It reversibly catalyzes the transfer of an acyl group from acyl-CoA to L-carnitine, playing a crucial role in transporting fatty acids across the inner mitochondrial membrane.
Carnitine palmitoyltransferase I (CPT-I) catalyzes the reaction between carnitine and fatty acyl-CoA, with or without malonyl-CoA, to form acylcarnitine and release coenzyme A (CoA-SH). The released CoA-SH reacts with Ellman's reagent (DTNB) to produce yellow TNB. The CPT-I activity is then calculated by measuring the absorbance at 412 nm.
