Microbial biochemical reactions refer to chemical-based assays for detecting microbial metabolites. Biochemical tests are commonly used to identify microorganisms that are difficult to distinguish by morphology and other characteristics. Accordingly, microbial biochemical reactions serve as one of the critical foundations for the taxonomic identification of microorganisms. Common biochemical tests include assays for carbohydrate metabolism, amino-acid and protein metabolism, carbon-source and nitrogen-source utilization, enzyme-related metabolic tests, and so on.Certain bacteria produce catalase, which catalyzes the decomposition of hydrogen peroxide into water and nascent oxygen, subsequently forming oxygen gas and producing bubbles. The catalase test reagent is formulated with oxidants, deionized water and other ingredients and is mildly corrosive. This product is intended for the catalase test of bacteria. For research-only use. Not suitable for clinical diagnosis or other applications.
Test Procedures (For Reference Only)
1. Method 1: Place 1-2 drops of catalase test reagent onto a clean glass slide. Pick one loop of test bacteria cultured on an agar slant and mix thoroughly with the reagent. Bubble formation indicates a positive catalase-test result; the absence of bubbles indicates a negative result.
2. Method 2: Pipette 2 mL of catalase test reagent into a clean small test-tube. Use a thin glass rod to collect the test strain and submerge it beneath the liquid surface of the reagent. A bubble-generating reaction is regarded as positive.
3. Method 3: Add 1 mL of catalase test reagent onto the culture (colony or bacterial lawn). Bubble formation indicates a positive reaction.
Precautions
1. Do not culture test organisms on blood-containing media, as this may cause false-positive results.
2. Platinum loops cannot replace glass rods. Contact between platinum loops and the reagent may produce false bubbles.
3. Set up control groups for each reaction. Staphylococcus aureus is used as the positive-control strain, and Streptococcus spp. as the negative-control strain.
4. Store the reagent in a tightly-sealed container. The shelf-life will be greatly shortened once opened.
5. For your personal safety, wear a lab coat and disposable gloves during operation.
6. Use the reagent promptly after opening to avoid compromised experimental performance.
