CMP–sialic acid synthetase (NmCSS), EC 2.7.7.43, derived from Neisseria meningitidis. This enzyme activates sialic acid to produce CMP-sialic acid, which serves as the sialic acid donor for glycosyltransferases, and acts as a tool enzyme for sialylation transfer reactions. This product is recombinantly expressed in Escherichia coli, featuring high purity and excellent compatibility, and is applicable in one-pot multi-enzyme systems.

Instructions for Use
Add substrates (Neu5Ac and CTP) and 5 mM MgCl₂ to the reaction system; appropriate buffer salts (e.g., Tris-HCl) may be supplemented to adjust the pH to 7.5–8.5, followed by the appropriate amount of NmCSS according to reaction requirements. Addition of inorganic pyrophosphatase to the system can improve conversion efficiency.
Mix thoroughly and incubate the sample at 37 °C for 30–60 min.
Notes
When scaling up the reaction system, incubation time or enzyme dosage shall be adjusted according to practical conditions; low-speed shaking mixing is recommended.
Most common biological buffers such as Tris and PBS are compatible. Buffers outside this pH range (e.g., acetate buffer) may also be usable, but incubation time and enzyme dosage need to be optimized on a case-by-case basis.
