Dehydroascorbate reductase catalyzes the reduction of dehydroascorbic acid (DHA) by reduced glutathione (GSH) to generate ascorbic acid (AsA). Reduced glutathione reacts with 5,5′-dithio-bis-(2-nitrobenzoic acid) (DTNB) to produce 2-nitro-5-mercaptobenzoic acid (TNB) and glutathione disulfide (GSSG). TNB exhibits a characteristic absorption peak at 412 nm. Therefore, the activity of dehydroascorbate reductase can be determined by measuring the rate of decrease in reduced glutathione.
