Overview

  Diff-Quik Staining is a rapid staining method modified from Wright Staining and is one of the commonly used staining techniques in cytological examinations. The staining solution is formulated based on the rapid staining method recommended by the World Health Organization (WHO). Similar to Wright Stain, it is derived from the Romanowsky Stain technique. The staining results are also very similar to those of Wright's stain, but Diff-Quik staining requires an extremely short time—typically completed within 90 seconds.

  The Diff-Quik Staining Solution contains a fixative and is primarily used for staining blood cell smears, bone marrow smears, vaginal secretion smears, exfoliative cell smears, etc. It is particularly suitable for batch immersion staining and provides a clear background without precipitate. This staining solution is intended for research use only and should not be used for clinical diagnosis or other purposes.

Instructions for Use

1. Prepare blood or bone marrow smears using standard methods. After air-drying or drying over an alcohol lamp flame, fix with Diff-Quik Fixative for 20–30 seconds.

   Note: The fixation strategy varies depending on the sample. Please refer to Notes 3–5 for details.

2. Stain with Diff-Quik Solution Ⅰ for 5–10 seconds.

3. Stain with Diff-Quik Solution Ⅱ for 10–20 seconds.

4. Rinse with water and immediately examine under a microscope while wet.

5. For slides deemed valuable after observation, return them to the lab for clearing with xylene, followed by mounting and preservation.

Precautions

1. Blood or bone marrow smears should be of uniform thickness to avoid affecting staining quality.

2. For blood cell smears, use fresh whole blood or EDTA-anticoagulated blood. The blood film must be completely dry; otherwise, cells may detach during staining.

3. For bone marrow smears, air-dry prepared slides quickly by waving or fanning to prevent cell shrinkage, deformation, or hemolysis due to humidity. Do not dry using high heat or flame.

4. For vaginal secretion smears, fix fresh specimens promptly with flame or alcohol to prevent cell deformation.

5. For exfoliative cell smears, fixation can be achieved by air-drying or using a fixative solution.

6. During staining, do not pour off the stain first or rinse the slide forcefully, as this may cause dye precipitation on the smear.

7. Staining solutions can be reused but not excessively. Filter if precipitates form.

8. Over-stained slides can be decolorized with methanol or alcohol. Re-staining is not recommended.

9. Adjust staining time or working solution concentration if results are too dark or too light.

10. pH affects staining. Ensure slides are clean and free from acid/alkali contamination.

11. Wear a lab coat and disposable gloves for safety.
