The purpose of fixation is to preserve the original morphological structure of cells and tissues. Fixatives prevent autolysis of tissues and cells by endogenous lysosomal enzymes and inhibit the growth of bacteria and molds. Fixatives are classified into aldehyde‑based, mercury‑based, alcoholic, oxidizing‑agent and picrate fixatives. Formalin (aldehyde‑based) and ethanol (alcohol‑based) are among the most commonly‑used types.
The environmentally‑friendly tissue fixative (concentrated) is mainly formulated with plant polysaccharides and polyvinyl alcohol. It contains no harmful substances such as formaldehyde and is a non‑toxic eco‑friendly fixative. It shall be mixed with 3 volumes of 95% ethanol before use. It produces well‑defined morphological structures and high background contrast for conventional paraffin‑embedded sections. It exhibits better performance than formaldehyde‑based fixatives in immunohistochemistry and molecular pathology. Most antibodies do not require antigen retrieval, which reduces fixation‑induced false‑positive results. Specimens fixed with this fixative show histomorphology comparable to formaldehyde‑fixed samples, with clear tissue‑cellular architecture and no defects such as vacuolization or karyopyknosis. This fixative is colorless and free of irritating and toxic volatile components. Being formaldehyde‑free, it can fully replace formaldehyde as a routine fixative in histopathology and cytopathology laboratories, minimizing health hazards to personnel in pathology and surgery departments as well as environmental pollution. It is suitable for routine histopathological sectioning and staining, classical histocytochemistry, immunohistocytochemistry, in‑situ hybridization and other advanced molecular pathological techniques. Appropriate for fixation of most tissues and cells, it is one of the commonly‑used fixatives for immunohistochemistry and cultured‑cell samples, and can well preserve the morphological structure of tissues and cells as well as nucleic acids. Procedures (for reference only):
1. Follow specific experimental requirements. Generally, mix with 3 volumes of 95% ethanol prior to application.
2. Fixation time for general specimens: 1‑4 h per millimeter of tissue thickness; extend fixation duration for large‑size specimens.
3. Post‑fixed tissues can be stored long‑term in 10% formalin or 70% ethanol.
Precautions
1. Fixation efficacy may decline after long‑term storage once the container is opened.
2. Avoid excessive fixation duration, which may lead to over‑crosslinking of biomacromolecules; fixation time varies according to tissue thickness.
