Eosinophils are one type of inflammatory cellular component and mostly function in allergic diseases. The cytoplasm of eosinophils contains secondary granules, which mainly comprise basic proteins, such as eosinophil cationic protein, eosinophil-derived neurotoxin, and eosinophil peroxidase.
Eosinophil Staining Solution consists of Carbol 2R Staining Solution and Hematoxylin Staining Solution. The cytoplasm of eosinophils is stained pink, and the nucleus is stained blue. This reagent is for research use only and not for clinical diagnosis or other purposes.

Protocol (For Reference Only):
1. Tissue fixation (10% neutral formalin fixative is recommended), followed by routine dehydration, embedding, and dewaxing to water.
2. Stain in Carbol 2R Stain for 15–20 min, then rinse with distilled water.
3. Stain in Hematoxylin Stain for 3–8 min, then rinse with distilled water.
4. Dehydrate in graded ethanol, clear routinely, and mount with neutral balsam.
Staining Results:
Cytoplasm of eosinophils: Pink
Nuclei: Blue
Precautions:
1. If differentiation is unsatisfactory, 0.5% hydrochloric acid alcohol may be used for differentiation to remove background.
2. Can also be used for staining cell smears, but the staining time should be shortened accordingly; fix with 4% paraformaldehyde solution.
3. Please use the reagent as soon as possible after opening to avoid affecting experimental results.
