Product Introduction:
Currently, exosome preservation solutions available on the market have problems such as few types and poor effectiveness. They are likely to cause damage to the structure of exosomes during preservation, loss of biological activity or degradation of contents, which affects subsequent downstream research or applications such as exosome detection and labeling. To solve this problem, Aladdin has developed a preservation solution with simple components that does not affect the activity of exosomes. It can preserve exosomes for a long time at low temperatures, is suitable for the preservation of liquid and lyophilized forms, and can effectively reduce the negative impact of lyophilization on exosomes.

1.CD9 protein content of exosomes under different preservation conditions
ELISA protein quantification experiments were performed on exosomes under different preservation conditions. The results showed that compared with 25°C, the exosomes preserved at 4°C for one month had less protein content loss, and the loss rate in the preservation solution was significantly reduced .
2.Exosome particle loss rate
NTA detection was performed on exosomes under different preservation conditions. The results showed that when stored at room temperature and 37°C for one month, the loss rate of exosome particles was as high as more than 80%. The loss rate of exosome particles stored at 4°C, -20°C and -80°C for one month was significantly lower than that at room temperature and 37°C.
3.Morphology of exosomes under different preservation conditions
Compared with freshly isolated exosomes, the membrane structure of exosomes stored in PBS at 4°C for 14 days is incomplete, which will lead to the leakage of exosomal proteins and the dissociation of loosely bound "exosomal peripheral" proteins. When exosomes are stored in the preservation solution, lyophilized and then kept at -20°C for 3 months, electron microscopy results show that the exosomal membrane structure is intact, and the morphology is complete and clear.
Operating Procedures:
1.Take 20μL of EV Save Buffer (2×) and add 20μL of ddH₂O to dilute it to 1×.
2.Take 10¹¹ exosomes as an example, add 40μL of EV Save Buffer (1×), mix well and store.
Notes and Disclaimer:
1.If exosomes need to be used within a short period of time, store them at 4°C to avoid the impact of freezing and thawing on exosomes.
2.After adding exosomes to this preservation solution, they can be stored at 4°C for 6 months, and can be stably preserved at -80°C after lyophilization.
