Principle
Suitable for partial examination of nematodes, tapeworm eggs and some protozoan encystments. Can be divided into saturated saline floatation method, zinc sulfate centrifugal floatation method and sucrose centrifugal floatation method. Move 1. (1) Pick soybean-sized feces (about 1 gram) with a bamboo stick and place it in a floating cup.
(2) Mash the feces and stir it well, then add saturated salt water until it is slightly higher than the mouth of the cup but does not overflow.
(3) Take a clean slide and put it over the mouth of the tube without producing air bubbles when in contact with the liquid. After resting for 15 minutes, lift the slide flat, turn the slide over quickly and examine under the microscope.
2、Zinc sulfate centrifugal floatation method
(1) Take about 1g of feces, add 10 times of water, and pour the fully stirred fecal liquid through a sieve into the centrifuge tube.
(2) Centrifuge at 2500 rpm for 1~2 minutes, pour off the upper liquid, then add water and centrifuge, repeat several times, until the supernatant is clarified.
(3) Pour off the supernatant, add a small amount of 33% zinc sulfate solution (specific gravity 1.18) to the sediment, mix well, then add 33% zinc sulfate solution to 1cm from the mouth of the tube, centrifugation for 1 minute and then stand still for a moment, take the liquid surface of the floating aggregates placed on the slide microscopic examination. If checking the encapsulation, need to add 1 drop of iodine solution staining after microscopic examination.
3、Sucrose centrifugation method
(1) Take about 5g of feces, add 15-20ml of water, and filter it through 260 mesh nylon bag or 4 layers of gauze.
(2) Take the filtrate centrifugation for 5~10 minutes, discard the supernatant, add sucrose solution (sucrose 500g, distilled water 320ml, carbolic acid 6.5ml) and then centrifugation, and then as saturated saline flotation method, take the surface liquid membrane microscopy (high power or oil microscope).
