1. Staining: HE

2. Visualization by the naked eye:

The specimen is a transverse section of small intestine; the lumen of the tube is lined with many tiny finger-like projections, the surface of which is where the epithelial tissue to be observed is located.

3. low magnification:

Find the luminal surface of the small intestine, see many intestinal villi of different lengths, covered with a single layer of columnar epithelium. Choose a vertical section with clear structure, move to the center of the field of view, and turn to high magnification observation.

4. High magnification observation:

The epithelial cells were high columnar, closely and neatly arranged. The nucleus was oval and stained purple-blue; it was located at the base of the cell; the cytoplasm was stained pink. The basal surface of the epithelium is attached to connective tissue. In a typical vertical section, the nuclei of neighboring columnar cells are seen to be at approximately the same height, and the nuclei are arranged in a single row throughout the epithelium. There were many vacuolated cup-shaped cells sandwiched between the columnar cells, and the mucogenic granules in the upper part of the cells were dissolved during the preparation process, so the cells were vacuolated; the nuclei of the cup-shaped cells were mostly triangular in shape, and the staining was deeply located in the basal part of the cells.
