Universal Fixing Solution is a fixing solution used for film processing after exposure in Western, Northern, Southern blot and other related experiments. This formula is specially optimized for X-ray film fixing, and it can also be applied to conventional black-and-white negatives and photographic paper. Universal Fixing Powder is the powdered form of Universal Fixing Solution; it can be prepared into working fixing solution after dissolving in deionized water. This reagent is intended for research use only and shall not be applied to clinical diagnosis or other purposes.
Materials to Be Prepared by User
1. X-ray films, red safe light, magnetic stirrer, beaker
2. Tap water, deionized water, developing solution
Operating Procedures (Including Development & Fixation, For Reference Only)
1. Prepare Universal Fixing Solution: Add approximately 800 mL deionized water (around 25℃) into a 1 L beaker, add the entire pack of Universal Fixing Powder and stir thoroughly until fully dissolved, then top up with deionized water to 1000 mL for use. Filtration with a 100-mesh filter screen is optional if necessary.
2. Development: Fully immerse the X-ray film in developing solution for 10 seconds to 2 minutes generally. Adjust the developing time according to the actual developing effect. Observe under red safe light and terminate development once the expected band intensity is achieved, then proceed to the next steps.
3. Rinsing: Rinse the film with tap water for 2-3 minutes to completely remove residual developing solution.
4. Fixation: Submerge the film in fixing solution for about 2-5 minutes. Extending the fixation time to 10 minutes will not produce obvious adverse effects on the final result.
5. Final Washing: Rinse thoroughly with tap water for 2-5 minutes to eliminate residual reagents.
Precautions
1. All procedures including exposure, development and fixation must be performed in a strictly light-proof environment.
2. For film development: 2-6 minutes for roll film developing, 1-3 minutes for print making.
3. For film fixation: around 10 minutes for negative film, 10-15 minutes for print making.
4. The working solution can be reused for 5-10 times, with a maximum reuse limit of 20 times; adjust the service life based on practical performance.
5. After preparation into liquid form, store the solution away from light at 4℃, valid for 6 months.
6. Wear a lab coat and disposable gloves during operation for personal safety and health.
7. Use the reagent as soon as possible after opening the package to avoid compromised experimental performance.
