Yeasts are unicellular eukaryotic microorganisms with distinct differentiation between cell nucleus and cytoplasm. Yeast cells are generally oval, spherical, cylindrical or lemon-shaped, and each strain has a specific morphological size. Attention should be paid to cell morphology when observing individual yeast cells.One or several transparent tiny droplets known as vacuoles exist in the cytoplasm of yeast cells. Vacuoles of vigorously growing yeast contain no inclusions, while vacuoles in aged cells accumulate granular storage substances such as fat droplets and glycogen granules. Vacuoles can be stained red with neutral red; fat droplets can be stained blue-black with Sudan Black; glycogen granules can be stained dark reddish-brown with iodine solution. These special structures inside yeast cells can then be observed under an optical microscope.

1% Glycogen Granule Staining Solution is mainly composed of potassium iodide and 1% iodine. It is applied to stain yeast smears, staining glycogen granules inside yeast cells deep red or reddish-brown. This reagent is intended solely for scientific research and shall not be used for clinical diagnosis or other purposes.

Operating Procedures (For Reference Only)：

1. Prepare a smear of Saccharomyces cerevisiae and allow it to air-dry naturally.

2. Drop 1~2 drops of this staining solution onto the smear and stain for 1~2 minutes.

3. Place a coverslip over the sample and observe the staining result under a microscope.

Precautions：

1. Mark a circle on the reverse side of the slide before preparing the smear to locate the observation area in follow-up experiments.

2. This product is restricted to scientific research performed by professional personnel. It must not be used for clinical diagnosis or treatment, food or pharmaceutical production, nor stored in residential premises.

3. Wear a lab coat and disposable gloves during operation to ensure personal safety and health.
