Pectin is a major component of the primary cell wall and middle lamella, primarily composed of protopectin, pectin methyl ester, and pectic acid. It contains monosaccharide components such as galacturonic acid, lactose, arabinose, and glucuronic acid. It is the most abundant polysaccharide in the cell walls of many higher plants, and its unique physicochemical properties directly affect the texture and quality of plant‑derived foods. Pectin molecules are cross‑linked through Ca²⁺ bridges and other ionic bonds, hydrogen bonds, glycosidic bonds, ester bonds, and benzene ring couplings. Depending on the extraction method used, pectin can be classified into water‑soluble pectin (WSP), ionically bound pectin (ISP), and covalently bound pectin (CSP). This kit employs an acid solution containing a chelating agent to extract ionically bound pectin (ISP), and measures it using the carbazole colorimetric method. The principle is as follows: Pectin is hydrolyzed to galacturonic acid, which then undergoes a condensation reaction with carbazole reagent in sulfuric acid solution to form a colored product with a maximum absorption peak at 530 nm. By measuring the absorbance, the ISP content in the sample can be quantitatively calculated.
