Overview

Non-specific binding to Leukocytes is a well-known challenge in flow cytometry. Though the specific mechanism of non-specific binding is not known, the binding of cyanine acceptor fluorophores to CD64 has been implicated in the literature. Mouse Fc Receptor Blocking Solution Plus not only specifically blocks non-specific antibody binding to CD16/CD32 (low-affinity IgG Fc receptors III and II), preventing non-specific binding of immunoglobulins to Fc receptors, but also effectively inhibits non-specific staining by various PE, PerCP, and APC tandem dyes on leukocytes. Compared to using anti-mouse CD16/32 mouse Fc receptor blocker alone, the new product demonstrates superior blocking performance against non-specific signals on Cyanine dyes.

This buffer contains specialized Rat IgG containing 0.05% ProClin 300. It is not recommended to be used for staining Rat IgG. Handle as biohazard agent.

Mouse Fc Receptor Blocking Solution Plus blocks both clone P90A23 and 2.4G2 also raised against mouse CD16/32.

Mouse Fc Receptor Blocking Solution Plus (M1374079) reduces nonspecific staining but preserves specific staining of mouse bone marrow cells (BMC) for CD11b. Mouse BMC were either not preincubated (Left Plots) or were preincubated (Middle Plots) with Mouse FcR Blocking Reagent (Ab219990), or were preincubated (Right Plots) with Mouse Fc Receptor Blocking Solution Plus as indicated. The cells were then stained with Rat IgG (PE-Cy5) (Ab190072) and recombinant CD11b Antibody (AF405) (Ab213132). Two-dimensional pseudocolor plots of CD11b vs Rat IgG show that the Mouse Fc Receptor Blocking Solution Plus reduces non-specific binding compared to staining without any blocking reagent and with Mouse FcR Blocking Reagent.

The cells were then stained with Rat IgG (PE-Cy5) (Ab190072) and CD45R Rat mAb (FITC) (Ab191667). Two-dimensional pseudocolor plots of CD45R vs Rat IgG show that the Mouse Fc Receptor Blocking Solution Plus reduces non-specific binding compared to staining without any blocking reagent and with Mouse FcR Blocking Reagent.
