Penicillin G Acylase (PGA; EC 3.5.1.11), also known as penicillin acylase, penicillin amidase or penicillin amino hydrolase, is a hydrolase that acts on the side chain of penicillin G.Penicillin G acylase acts on penicillin G, cephalosporin G and related antibiotics to produce β-lactam antibiotic intermediates 6-aminopenicillanic acid (6-APA) or 7-amino-desacetoxycephalosporanic acid (7-ADCA) and a by-product phenylacetic acid (PAA). These antibiotic intermediates are structural units of semi-synthetic penicillin antibiotics (ampicillin, amoxicillin, cloxacillin, sulbactam, etc.) and cephalosporin antibiotics (cefadroxil, cephalexin, etc.).
This enzyme is a heterotetrameric protein composed of 2 α-subunits and 2 β-subunits derived from a single polypeptide chain precursor. PGA is a member of the N-terminal nucleophile hydrolase superfamily, which shares a common fold structure at or near the active site. The N-terminal position of the active site carries a catalytically active serine, cysteine or threonine residue.
