Overview
The fixative stabilizes the cell membrane and preserves the binding of antibodies to antigens on the cell surface. The Permeabilization Buffer creates pores in the fluid, intact cell membrane to allow antibodies to enter the cell. For the detection of intracellular antigens and DNA by flow cytometry, cells are fixed and permeabilized, then incubated with the corresponding antibodies or detection reagents before analysis.
Instructions for Use:
Harvest the cells to be tested and centrifuge at 1000 rpm for 10 minutes. Discard the supernatant.
Add 500 µL of pre-chilled (4°C) fixative.
Incubate at room temperature for 10 minutes.
Centrifuge at 1000 rpm for 10 minutes.
Discard the supernatant. Wash the cells once with PBS, then centrifuge at 1000 rpm for 10 minutes.
Discard the supernatant. Add 1.5 mL of permeabilization agent.
Incubate at room temperature for 10-15 minutes.
Centrifuge at 1000 rpm for 5 minutes.
Discard the supernatant. Resuspend the cells in an appropriate volume of permeabilization agent.
Add the detection antibody and stain for 15 minutes.
Wash the cells once with PBS and centrifuge at 1000 rpm for 5 minutes.
Discard the supernatant. Resuspend the cells in 200 µL of PBS for analysis on the flow cytometer.
Precautions
The Permeabilization Buffer contains 0.05% ProClin 300. Avoid contact with skin, eyes, and mucous membranes during use.
