Phospholipase C (PLC, EC 3.1.4.3) is a lipid hydrolase that specifically hydrolyzes the glycerophosphoester bond at the C3 position of glycerophospholipids. It is widely distributed in microorganisms as well as in animal and plant tissues and cells, playing crucial roles in cellular metabolism, signal transduction, growth, and development.
Phospholipase C catalyzes the hydrolysis of O-(4-nitrophenyl)choline (NPPC) to produce p-nitrophenol (PNP), which exhibits a maximum absorption peak at 405 nm. PLC enzyme activity can be determined by measuring the increase rate of PNP at 405 nm.
