Phospholipase D (EC 3.1.4.4), also known as phosphatidylcholine hydrolase, is a general term for a class of enzymes that catalyze the hydrolysis of phosphodiester bonds and base exchange reactions. It is widely present in various organisms such as higher plants, animals, and bacteria, and performs physiological functions including involvement in cellular lipid metabolism, signal transduction, membrane formation, and stress resistance.
Phospholipase D catalyzes the hydrolysis of the substrate O-(4-nitrophenyl)choline (NPPC). With the addition of exogenous acid phosphatase, this reaction produces p-nitrophenol (PNP), which has a maximum absorption peak at 405 nm. The PLD enzyme activity can be determined by measuring the increase rate of PNP at 405 nm.
