Polygalacturonase (EC 3.2.1.15) is a cell wall-bound protein that catalyzes the cleavage of α-(1,4)-polygalacturonic acid in pectin molecules, participating in pectin degradation and leading to the disintegration of cell wall structure, which results in fruit softening. It is associated with fruit ripening, leaf and flower abscission, pathogen defense, cell expansion and development, as well as lignification, and holds significant research value in the fields of plant disease resistance and food storage and preservation.
Determination Principle: Polygalacturonase hydrolyzes pectic acid to produce galacturonic acid, which contains a reducing aldehyde group. This reacts with the DNS reagent to form a reddish-brown compound with a characteristic absorption peak at 540 nm. Polygalacturonase activity can be calculated by measuring the change in absorbance at 540 nm.
