Proline dehydrogenase (PDH, EC 1.5.1.2) catalyzes the reaction that converts proline to Δ1-pyrroline-5-carboxylate, serving as the rate-limiting step in the proline degradation pathway. Proline is the most widely distributed osmoregulatory substance; under stress conditions, many plants accumulate substantial amounts of proline by enhancing its synthesis and reducing its degradation. Decreasing PDH activity is of great significance for preventing osmotic stress damage to plants and protecting cellular structure.
Proline dehydrogenase (PDH) catalyzes the dehydrogenation of proline while reducing NAD⁺ to NADH. The PDH enzyme activity can be determined by measuring the increase rate of NADH at 340 nm.
