This series of Cell Cryopreservation Medium is developed in layers to meet diverse research demands, covering three product models: C1525335, C1525336 and C1525338. It covers three application scenarios including cost-effective routine cell culture, high-standard serum-free protein-based cryopreservation and dedicated low-temperature storage for red blood cells, compatible with most mainstream in vitro experimental samples in laboratories.

Instructions for Use: The freezing medium has been sterilized before filling. Warm it up to above 25℃ before use and operate in accordance with the cryopreservation protocol for red blood cells.
Cell Cryopreservation Protocol:
1. Pre-cryopreservation Preparation
(1) Prepare packed red blood cells (The volume of packed red blood cells prepared from 200 mL whole blood is approximately 68 mL).
(2) Place red blood cells and freezing medium at room temperature for 1 hour to bring both to a temperature of at least 25℃. This step may be omitted if red blood cells are isolated directly from whole blood kept at room temperature.
(3) Calculation of required freezing medium volume:
2.35 mL freezing medium is required per milliliter of packed red blood cells.
Volume of required freezing medium = Volume of packed red blood cells×2.35
Example: The packed red blood cells derived from 200 mL whole blood measure about 68 mL, so the required freezing medium volume is calculated as 68×2.35=160 mL.
2. Cryopreservation Procedures
(1) Transfer the packed red blood cells to be cryopreserved into a sterile container and record the cell volume as V₁.
(2) Calculate the required freezing medium volume as V₂= 2.35 mL×V₁.
(3) Add freezing medium equivalent to 1/4 of V₂ to the red blood cells, mix gently by pipetting, shaking or using a shaker, then incubate at room temperature for 30 min.
(4) Repeat step (3), adding 1/4 V₂ freezing medium each time. After all medium is fully added, incubate the mixture at room temperature for another 30 min.
(5) Transfer the red blood cell suspension into cryobags or cryovials.
(6) Place the samples into a cryobox and store in a -80℃ refrigerator (liquid nitrogen transfer is not required).
Cell Thawing Protocol:
1. Take the cryopreserved red blood cells out of the -80℃ refrigerator.
2. Immediately immerse them in a 37℃ water bath and shake gently until completely thawed; this process takes approximately 10 minutes.
3.Perform red blood cell washing right after thawing.
General Precautions:
1.Transfer the product to a 4℃refrigerator immediately after receiving.
2.In case of skin or eye contact during use, rinse with plenty of clean water at once, and seek medical treatment if discomfort occurs.
3.Do not use the product if turbidity, precipitation or other deterioration appears, and contact technical personnel immediately.
4.Open the bottle cap inside a biosafety cabinet to avoid cell contamination.
