Clotting time is defined as the time required for venous blood to fully coagulate after being drawn from the body. It serves as a screening assay reflecting the activity of various coagulation factors in the intrinsic coagulation pathway. Under the action of thrombin, fibrinogen in the test plasma is converted into fibrin. When anticoagulants increase in the test plasma, the thrombin time will be prolonged accordingly.
The Thrombin Time (TT) Assay Kit is used to determine the thrombin time of human and animal plasma. Prolonged thrombin time is observed in cases of elevated heparin or heparin-like anticoagulants, increased fibrin(ogen) degradation products (FDP), and hypo(a)fibrinogenemia. Shortened thrombin time may occur when microclots exist in blood samples or calcium ions are present. This kit is intended for research purposes only and shall not be used for clinical diagnosis or other applications.

Materials to be prepared by users
1)Sodium citrate anticoagulant blood collection tubes, blood collection needles, tourniquets
2)Centrifuge, timer or stopwatch, water bath
Operating Procedures (For reference only)
1)Preparation of test plasma: Collect 1.8 mL fresh venous blood sample, mix gently with 0.2 mL sodium citrate anticoagulant (109 mM) at a volume ratio of 9:1 with gentle inversion. (Alternatively, collect blood using plastic tubes or siliconized glass tubes pre-filled with 1/10 volume of sodium citrate anticoagulant.)
2)Centrifuge at 3000 rpm (or 2500 g) for 10~15 min. Collect the supernatant (platelet-poor plasma) and transfer it into plastic test tubes or centrifuge tubes to avoid platelet activation. Normal control plasma shall be set up simultaneously.
3)Pipette 0.1 mL anticoagulated test plasma and incubate in a 37 °C water bath for 5 min.
4)Add 0.1 mL thrombin solution and record the clotting time. Repeat the test 2-3 times and calculate the average value.
Reference Range
14.0~21.0 s. Results exceeding the normal control value by more than 4 s are considered abnormal.
Precautions
1)A normal control shall be tested in parallel for all operations described above.
2)Samples should be protected from hemolysis and tissue fluid contamination.
3)Assays should be performed promptly after sample collection, preferably within 1 h. Storage in a refrigerator must not exceed 4 h.
4)Disposable plastic syringes or siliconized glass syringes must be used for blood collection; plastic tubes or siliconized glass tubes are required for blood storage. Ordinary glass containers are not recommended to prevent activation of coagulation factors.
5)The tourniquet should not be tied too tightly during blood collection, and the constriction time should not exceed 5 minutes; otherwise, coagulation and fibrinolytic factors may become activated.
6)Centrifuge samples at 3000 rpm for 10–15 min for plasma separation to ensure complete removal of platelets.
7)Heparin, EDTA or oxalate should not be used as anticoagulants in this assay.
8)The testing temperature should be maintained at 36.5~38.5 °C. Temperatures below or above this range can prolong TT values.
9)This product is for research purposes only and must not be used for clinical diagnosis or other applications.
10)Wear a lab coat and disposable gloves during operation for your personal safety and health.
