Tissue‑cell dissociation solution is applied for tissue dissociation to prepare single‑cell suspensions in in‑vitro tissue culture and primary cell culture, as well as for detachment of adherent cells during subculture. Composed of trypsin, EDTA, phenol red and other components, it hydrolyzes intercellular proteins such as extracellular matrix, dissociating tissues or adherent cells into single‑cell suspensions for downstream experiments.

Sterilized by filtration, Trypsin Cell Dissociation Solution is ready‑to‑use for dissociation of cultured cells and tissues. It features convenient operation, fast processing, high stability, good safety and favorable cell viability. Most adherent cells can be dissociated within approximately 2 min at room temperature. Phenol red serves as a pH indicator.

Instructions for Use

1. Dissociation of adherent cells

1.1 Remove culture medium. Rinse cells once with sterile PBS, Hanks’ balanced salt solution or serum‑free medium to eliminate residual serum.

1.2 Add a minimal volume of trypsin dissociation solution to just cover the cell monolayer. Incubate at room temperature for 1‑2 min. Incubation time varies among cell types; extend incubation appropriately for strongly adherent cells.

1.3 Monitor under microscope: stop when cells exhibit obvious shrinkage, morphological alteration at the bottom of culture vessels, or can be gently dislodged by pipetting. Remove dissociation solution. Add serum‑supplemented complete medium and pipette to harvest cells for subsequent experiments.

1.4 If dissociation is insufficient, add fresh trypsin solution and repeat digestion.

1.5 If over‑digestion occurs and partial cells detach before pipetting, pipette to harvest all cells directly in trypsin solution. Centrifuge at 1000‑2000 g for 1 min to pellet cells. Remove trypsin supernatant as much as possible, then resuspend cell pellet in serum‑containing complete medium for downstream use.

2. Tissue dissociation

Digestion time varies greatly for different tissues. Terminate digestion when tissues can be fully triturated.

Precautions

1. Optimal digestion time should be determined empirically according to specific tissue or cell types. Over‑digestion impairs cell attachment and proliferation.

2. This product contains no bacteriostatic agents. Strict aseptic manipulation is required to prevent microbial contamination.

3. Do not store long‑term at 4 °C. Avoid repeated freeze‑thaw cycles. Aliquot for long‑term storage.

4. Wear laboratory coat and disposable gloves for personal protection during handling.
