Übersicht

Xylan is a major component of plant cell walls and is also an abundant and utilizable hemicellulose. Xylanase refers to a group of enzymes that specifically degrade hemicellulose and xylan into xylooligosaccharides and xylose. Xylanases are mainly derived from bacteria and fungi, including anaerobic bacteria, aerobic bacteria, mesophilic microorganisms, thermophilic microorganisms, and extremophiles. Xylanases are mainly classified into three types: ① β-1,4-D-endoxylanase (EC 3.2.1.8), which cleaves β-1,4 glycosidic bonds from within the main chain of xylan, rapidly reducing the viscosity of the xylan solution; ② β-1,4-D-exoxylanase (EC 3.2.1.92), which acts on the non-reducing ends of xylan, cleaving individual xylose units and continuously increasing the reducing power of the reaction system; and ③ β-xylosidase (EC 3.2.1.37), which cleaves xylooligosaccharides, yielding mainly xylose, xylobiose, xylotriose, and other xylooligosaccharides as hydrolysis products. Based on their physicochemical properties, xylanases can also be divided into two main categories: alkaline xylanases with a molecular weight of less than 30 kDa and acidic xylanases with a molecular weight greater than 30 kDa. Generally, bacteria can produce both acid and alkaline xylanases simultaneously, while fungi typically only produce low-molecular-weight alkaline xylanases. Essentially, all xylanases possess both exo-enzyme and endo-enzyme activities, differing only in their relative activity levels. As an industrial enzyme preparation, xylanase has significant potential applications. Firstly, xylanase can convert xylan from agricultural and wood industry waste into xylose. Secondly, researchers such as Wong have suggested that xylanase can be used in processes like juice clarification, and in the extraction and processing of coffee, vegetable oils, and starch. The detection principle of this kit is as follows: Xylanase degrades xylan into reducing oligosaccharides and monosaccharides. Under boiling water bath conditions, these products can react with 3,5-dinitrosalicylic acid (DNS) reagent to produce a color reaction. The intensity of the reaction solution's color is directly proportional to the amount of reducing sugars produced by the enzymatic hydrolysis, which in turn is directly proportional to the xylanase activity in the reaction solution. Therefore, by measuring the color intensity of the reaction solution, the xylanase activity can be calculated. This product is intended for research purposes only and is not suitable for clinical diagnosis or other uses.
