Cellular agglutination reactions and cell membrane permeability

Summary

Cell membranes are unit membranes composed of lipid bilayers studded with proteins, which in turn bind to sugar molecules as a branched sugar coat on the cell surface. Lectins are a class of proteins that reversibly bind specific sugar groups and have the ability to cause cell agglutination, blood group identification, promotion of lymphocyte division, and pest control and insect resistance. Lectins make cells agglutinate because they are connected to the oligosaccharide chains of the cell coat, playing the role of "bridge".

I. Cellular agglutination reaction1. 2% duck blood erythrocyte suspension preparation: fresh duck blood plus acd anticoagulant, saline wash 5 times, each time 2 000 r/min, centrifugation for 5 min, according to the erythrocyte pressure volume with saline to form 2% duck blood erythrocyte suspension.2. Potato lectin preparation: 2 g of potato, add 10 ml of PBS buffer and soak for 2 h. The potatoes should be soaked for 2 hours.3. Cellular agglutinationPotato agglutinin 1 drop2% duck blood erythrocyte solution 1 dropMix well on the slide, let stand for 20 min, microscopic examination

II. Permeability of cell membranes
1. Hypotonic solutionsOne test tube:1 ml of duck blood erythrocyte fluidddh2o 10mlShake gently and mix well, observe the color change, if there is hemolysis, note down the time since the addition of duck blood until the solution is clear and clarified (pay attention to the combination of microscopic examination).
2. Isotonic solutions0.17 mol/l sodium chloride, 0.17 mol/l ammonium chloride, 0.17 mol/l ammonium acetate, 0.17 mol/l ammonium nitrate, 0.12 mol/l ammonium oxalate, 0.12 mol/l ammonium sulphate, 0.32 mol/l glucose, 0.32 mol/l glycerol, 0.32 mol/l ethanol, 0.32 mol/l acetone, etc. The same experiment is carried out with eight solutions. solutions were subjected to the same experiment with the same steps as above.
