Z-DEVD-AMC is a classic Caspase-3/7 substrate, used to detect the enzymatic activity of Caspase-3/7 in vitro or in cell lysis systems. This molecule consists of a tetrapeptide sequence DEVD (a specific recognition sequence for Caspase-3/7) and a fluorophore AMC (7-amino-4-methylcoumarin). Under normal circumstances, AMC is blocked by the DEVD sequence and does not emit light; when Caspase-3 or -7 is activated, it will recognize and cleave the bond between DEVD and AMC, releasing free AMC molecules, which then emit detectable blue fluorescence (Ex/Em ≈ 350/450 nm).
Z-DEVD-AMC is often used in microplate fluorescent enzyme activity assays, studies on drug-induced apoptosis, high-throughput drug screening systems, etc., and is particularly suitable for Caspase activity curve analysis and time-dependent experiments. Its advantages include fast reaction speed, strong signal, and low background. However, due to its relatively short excitation wavelength (in the UV region), it is recommended to use a fluorescence microplate reader or fluorescence spectrophotometer equipped with an ultraviolet excitation channel. In addition, it can also be used for fluorescent quantitative detection after cell lysis, but is not suitable for direct imaging of living cells. During operation, attention should be paid to avoiding strong light exposure to reduce the self-luminescence attenuation of AMC.
