The purpose of fixation is to preserve the original morphological structure of cells and tissues. Fixatives prevent autolysis of tissues and cells induced by endogenous lysosomal enzymes and inhibit the growth of bacteria and fungi. By coagulation and adduct formation, fixatives alter the internal structure of proteins to inactivate enzymes and induce physical changes to nuclei and extracellular components. Fixatives are mainly classified into aldehyde, mercury, alcohol, oxidant and picrate types, among which formalin (aldehyde type) and ethanol (alcohol type) are widely used.
AAF Fixative (for tender tissues), also named 50% FAA/AFA Fixative or universal fixative, consists of 50% ethanol (A), 5% acetic acid (A) and 5% formaldehyde (F). It is suitable for fixing young plant materials such as single plant cells and filamentous algae, but not for ordinary plant tissues. It effectively prevents tissue shrinkage. For aged or hard samples, 70% ethanol is recommended to replace 50% ethanol. FAA fixative can also be used as a preservative. This reagent is for research use only and not intended for clinical diagnosis.
Operating Procedures (For Reference Only)
1.Immerse fresh tissues in AAF Fixative (for tender tissues) and fix at room temperature for 1–12 hours.
2.If paraffin sections show poor staining due to insufficient fixation, re-fix the sections with AAF Fixative for 5 minutes before staining to significantly improve staining quality.
Precautions
1.AAF Fixative is harmful to the human body. Operate in a well-ventilated environment and avoid inhalation.
2.Fixation time varies with tissue thickness. Proper tissue cutting facilitates penetration of fixative. The optimal thickness of routine biopsy tissues is 2–4 mm, and shall not exceed 6 mm.
3.Use sufficient volume of fixative; the volume ratio of fixative to tissue block shall be greater than 10:1. Replace the fixative 1–3 times during fixation if the volume is insufficient.
4.Temperature significantly affects fixation. Elevated temperature accelerates fixation, yet excessive temperature should be avoided.
5.Fix fresh tissues immediately after dissection.
6.Use the reagent as soon as possible after opening to avoid compromised experimental results.
