Adenosine triphosphate (ATP) serves as the most fundamental carrier and the most direct source of energy in biological energy conversion. Determination of ATP content and calculation of energy charge can effectively reflect the energy metabolic status of organisms. This kit employs a coupled enzymatic method for ATP quantification: Under the combined catalysis of hexokinase (HK) and glucose-6-phosphate dehydrogenase (G6PDH), ATP hydrolysis drives the phosphorylation of glucose to glucose-6-phosphate, which is subsequently oxidized by G6PDH while reducing NADP⁺ to NADPH. By monitoring the increase in the characteristic absorbance peak of NADPH at 340 nm, under substrate-saturated conditions, the initial linear increase in OD₃₄₀ per unit time (ΔOD₃₄₀/min) is proportional to the ATP concentration. Accordingly, the ATP concentration in the sample can be precisely calculated.
