hemolytic test

Summary

This test uses the patient's erythrocytes to cause hemolysis to occur after incubation in their own serum (containing complement) at 37°C due to acid production from glucose decomposition that mellows the serum. This experiment is derived from the laboratory instruction of Mudanjiang Medical College undergraduate 5-year testing program.

Principle

This test uses the patient's red blood cells incubated in their own serum (containing complement) at 37°C to cause hemolysis to occur as a result of the breakdown of glucose to produce acid that mellows the serum.

Move

Take 2 ml of venous blood with a sterile dry syringe, remove the needle, slowly inject into a small sterilized test tube along the wall of the tube to avoid air bubbles, add a little sterilized paraffin to cover it, and place it in a 37℃ incubator to keep warm for 24 hours, and then take out the centrifugal precipitate.

Observe the color of the supernatant, such as hemolysis is positive, normal is negative.

Caveat

Note: The syringe should be dry, the small test tube should be clean, the operation process should avoid hemolysis or it will lead to false positives, after 24 hours in the incubator, if the white block has not retracted, use a small glass rod to gently separate it along the wall of the tube, and then centrifuge to observe the results.
