Overview
This kit employs an NADPH-coupled colorimetric method for the rapid determination of glycogen phosphorylase a (GPa) activity. GPa catalyzes the phosphorolysis of glycogen to generate glucose-1-phosphate, which, through the coupled reactions of phosphoglucomutase (PGM) and glucose-6-phosphate dehydrogenase (G6PDH), reduces NADP⁺ to NADPH. The generated NADPH reacts with a chromogenic reagent to form a colored product with a characteristic absorption peak at 450 nm. The rate of formation of this product is proportional to the GPa activity, thereby enabling the quantification of GPa activity in the sample. GPa is a key enzyme in glycogen breakdown, and its active form regulates glycogen metabolism.
