Overview
Hemicellulose is converted into reducing sugars upon acid treatment. The reducing sugars further react with 3,5-dinitrosalicylic acid (DNS) to form a brownish-red amino compound, which exhibits a characteristic absorbance peak at 540 nm. By measuring the change in absorbance at this wavelength, the hemicellulose content can be quantitatively determined.
