Overview

Pectin methylesterase (PE), also known as pectinesterase or pectin oxidase, is a member of the pectinase enzyme family. It catalyzes the hydrolysis of methoxy ester bonds in the pectin long chain, generating low-molecular-weight pectic acid and methanol, thereby increasing the solubility of pectin in water. PE is widely distributed in higher plants, as well as in bacteria and fungi capable of degrading cell walls. In plants, it plays a role in the endogenous regulation of cell wall and intercellular pectin content. The hydrolysis of pectin molecules catalyzed by PE releases H⁺, resulting in a decrease in the pH of the reaction system. By titrating with alkali to maintain the system pH constant at 7.8 (using phenolphthalein as the indicator to retain a pink color), the amount of NaOH consumed reflects the activity of pectin methylesterase.
