Overview

Adenosine triphosphate (ATP) is the most fundamental carrier and the most direct source of energy in biological energy conversion. The determination of ATP content and the calculation of the energy charge can effectively reflect the energy metabolism status of an organism. This kit employs a creatine kinase‑coupled phosphomolybdic acid colorimetric method for the determination of ATP content. The principle is as follows: Under the catalysis of creatine kinase (CK), ATP reacts with creatine to generate phosphocreatine and ADP. Phosphocreatine is further hydrolyzed to inorganic phosphate (Pi). Under acidic conditions, Pi reacts with ammonium molybdate to form a phosphomolybdic acid complex, which is then reduced by a reducing agent (such as ferrous sulfate or ascorbic acid) to generate a blue molybdenum blue product. This product exhibits a characteristic absorption peak at 700 nm. By monitoring the increase in absorbance at 700 nm under substrate‑saturating conditions, the initial linear increase in absorbance per unit time (ΔOD₇₀₀/min) is directly proportional to the ATP content, allowing for accurate calculation of the ATP concentration in the sample.
