Overview
Under the action of xanthine oxidase (XOD), xanthine and hypoxanthine generate hydrogen peroxide (H₂O₂). Peroxidase (POD) catalyzes the oxidation of 4‑aminoantipyrine by H₂O₂, which then undergoes a coupling reaction with TOOS to form a stable purple‑red quinone imine compound. This product exhibits a characteristic absorption peak at 550 nm. By measuring the change in absorbance at this wavelength, the content of xanthine and hypoxanthine in the sample can be quantitatively determined.
