Glucose‑6‑phosphatase (G6Pase, EC 3.1.3.9) is widely distributed in animals, plants, microorganisms, and cells. It is the rate‑limiting enzyme that catalyzes the hydrolysis of glucose‑6‑phosphate to glucose in the gluconeogenesis pathway, and plays a vital role in maintaining blood glucose homeostasis. This kit provides a convenient assay method suitable for the determination of G6Pase activity in various biological samples, including animal tissues, plant tissues, cells, serum, and plasma. The assay principle is as follows: G6Pase catalyzes the hydrolysis of glucose‑6‑phosphate to glucose. The generated glucose is then oxidized by glucose oxidase to produce hydrogen peroxide (H₂O₂). The H₂O₂ subsequently reacts through the Trinder reaction to generate a quinone imine compound, which exhibits a characteristic absorption peak at 554 nm. By measuring the rate of change in absorbance, the G6Pase activity can be reflected.
