Ponceau S, also named Tetrasodium 3-hydroxy-4-[2-sulfo-4-(4-sulfophenylazo)phenylazo]-2,7-naphthalenedisulfonate, has the molecular formula C₂₂H₁₂N₄Na₄O₁₃S₄, a molecular weight of 760.5688 and CAS No. 6226-79-5. It is a water-soluble anionic diazo dye synthesized by the coupling of diazoaminoazobenzene disulfonic acid and naphthol disulfonic acid into its tetrasodium salt. Negatively charged Ponceau S can bind to positively charged amino acid residues as well as the non-polar regions of proteins, producing distinct red protein bands. Protein staining with Ponceau S is reversible; the stain can be washed off with distilled water, PBS or other appropriate buffer solutions. This staining reagent is applicable for protein detection on PVDF membranes (Western blots), nitrocellulose membranes (NC) and cellulose acetate membranes, but is not compatible with nylon membranes.Ponceau S Staining Elution Solution, also referred to as destaining solution, is mainly composed of sodium hydroxide (NaOH) and used to elute Ponceau S stain from membranes. This product is for research use only and shall not be used for other purposes.
