Overview

Soluble starch synthase (SSS, EC 2.4.1.21) exists in a free form in the stroma of plastids and catalyzes the elongation of starch chains, being primarily responsible for the synthesis of amylopectin. SSS catalyzes the reaction of ADPG with a starch primer (glucan), transferring a glucosyl moiety to the primer and simultaneously generating ADP. A coupled enzymatic mixture in the reaction system sequentially reduces NADP⁺ to NADPH, and the amount of NADPH produced is directly proportional to the amount of ADP generated. Traditional methods determine SSS activity by measuring the increase in NADPH at 340 nm. However, these methods suffer from relatively low sensitivity and are prone to interference from pigments (e.g., green leaves) present in the samples. This kit provides a simple, sensitive, and rapid alternative: the NADPH generated from the enzymatic reaction reacts with a specific chromogenic probe to form a colored product. By measuring the rate of formation of this product at 450 nm, the SSS activity can be calculated.
