Overview

Glucokinase (GCK) is a key enzyme in the glucose metabolic pathway, participating in the first step of glycolysis by catalyzing the phosphorylation of glucose, which promotes insulin secretion and glucose metabolism, thereby effectively maintaining blood glucose homeostasis in vivo. GCK, also known as hexokinase IV, is primarily expressed in hepatocytes and pancreatic β‑cells. Compared to other hexokinases, GCK has a lower affinity for glucose (high Km). Therefore, activity measured under high glucose concentrations represents total hexokinase activity, while activity measured under low glucose concentrations represents high‑affinity hexokinases (such as HK I, II, and III). The difference between these two values corresponds to GCK activity. The assay principle of this kit is as follows: GCK catalyzes the substrate reaction to generate NADPH. NADPH then reduces WST‑8 to produce an orange‑yellow formazan, which exhibits a maximum absorption peak near 450 nm. By measuring the rate of increase in absorbance at 450 nm, the GCK activity in the sample can be calculated.
