Overview

Phospholipase D (PLD, EC 3.1.4.4), also known as phosphatidylcholine hydrolase, is a general term for a class of enzymes that catalyze the hydrolysis of phosphodiester bonds and base‑exchange reactions. It is widely distributed in higher plants, animals, bacteria, and other organisms, and participates in various physiological processes including cellular lipid metabolism, signal transduction, biofilm formation, and stress resistance. The assay principle of this kit is as follows: PLD catalyzes the hydrolysis of the terminal phosphodiester bond of phosphatidylcholine, generating phosphatidic acid and choline. Choline is then oxidized by choline oxidase to produce betaine and hydrogen peroxide (H₂O₂). In the presence of peroxidase (POD), H₂O₂ oxidizes 4‑aminoantipyrine and phenol to generate a pink quinone imine compound, which exhibits a characteristic absorption peak at 500 nm. By measuring the rate of change in absorbance at 500 nm, the PLD enzyme activity can be calculated.
