Overview

Anthocyanins are a class of natural pigments that are readily soluble in polar solvents and belong to the flavonoid compounds. They are widely distributed in the roots, stems, leaves, flowers, and fruits of plants, imparting a range of colors from red to purple, and are the primary pigments responsible for plant coloration. The pH differential method can be used to determine the anthocyanin content. At pH 1.0, anthocyanins exhibit a maximum absorption peak at 530 nm; whereas at pH 4.5, they are converted to the colorless chalcone form and show no absorption at 530 nm. Utilizing this property, the absorbance values at 530 nm and 700 nm are measured under the two pH conditions, respectively. The pH differential method can minimize the effects of variations in solution pH and solvent composition, and effectively eliminate interference from other non‑anthocyanin substances in the detection results.
