Proline dehydrogenase (ProDH) is a key enzyme located in the mitochondria that catalyzes the degradation of proline. Proline is one of the most widely distributed osmotic regulatory substances in plants. Under stress conditions, plants can accumulate large amounts of proline by enhancing its synthesis and inhibiting its degradation. Therefore, reducing ProDH activity is of great significance for regulating osmotic balance, alleviating osmotic stress‑induced damage to plants, scavenging free radicals, and protecting cellular structures. The assay principle of this kit is as follows: ProDH catalyzes the dehydrogenation of proline to generate Δ¹‑pyrroline‑5‑carboxylate (P5C). The released hydrogen is transferred via phenazine methosulfate (PMS) to reduce DCPIP (2,6‑dichlorophenolindophenol). DCPIP exhibits a characteristic absorption peak at 600 nm, and its rate of decrease in absorbance is positively correlated with ProDH activity. By measuring the rate of change in absorbance at 600 nm, the ProDH enzyme activity can be calculated.
