Pyruvate dehydrogenase (PDH, EC 1.2.4.1) is widely distributed in animals, plants, microorganisms, and cultured cells. It is the rate-limiting enzyme of the pyruvate dehydrogenase complex (PDHC), catalyzing the oxidative decarboxylation of pyruvate, and plays a critical role in linking glycolysis to the tricarboxylic acid (TCA) cycle. This kit employs an MTT colorimetric method for the determination of PDH activity: PDH catalyzes the decarboxylation of the substrate sodium pyruvate to generate hydroxyethyl-TPP, which subsequently transfers electrons to thiazolyl blue tetrazolium bromide (MTT). In the presence of the electron mediator phenazine methosulfate (PMS), MTT is reduced to yield a blue formazan product, which exhibits a characteristic absorbance peak at 566 nm. By continuously monitoring the initial linear increase in OD₅₆₆ per unit time (ΔOD₅₆₆/min) in kinetic mode, the PDH enzyme activity in the sample can be precisely calculated.
