Overview
Glutaminase (GLS, EC 3.5.1.2) is an amidase that catalyzes the hydrolysis of glutamine to glutamate and ammonia, playing an important regulatory role in nitrogen metabolism. Soil glutaminase (S‑GLS) catalyzes the hydrolysis of glutamine to L‑glutamate and ammonia (NH₃/NH₄⁺). This kit employs the indophenol blue colorimetric method for the determination of S‑GLS activity. Under strongly alkaline conditions, ammonia reacts with hypochlorite and phenol to form a stable, water‑soluble blue indophenol blue dye, which exhibits a characteristic absorption peak at 630 nm. By measuring the increase in absorbance per unit time, the S‑GLS enzyme activity in the soil sample can be quantitatively reflected.
