Alcohol dehydrogenase (ADH, EC 1.1.1.1) is widely present in many organisms. In humans and most animals, this enzyme is responsible for the detoxification of toxic alcohols. In yeast and certain bacteria, however, some alcohol dehydrogenases catalyze the reverse reaction as part of the fermentation process. This kit provides a rapid and sensitive method for the determination of ADH activity. The principle is as follows: ADH catalyzes the reaction of ethanol and NAD⁺ to produce acetaldehyde and NADH. The generated NADH further reacts with a specific chromogenic reagent, yielding a yellow product with a maximum absorption peak at 450 nm. By continuously monitoring the rate of formation of the yellow product at 450 nm, which is directly proportional to the ADH activity, the alcohol dehydrogenase activity can be quantitatively calculated.
