Overview

This kit employs an NADPH‑coupled chromogenic method in conjunction with a difference approach to separately determine the total glycogen phosphorylase activity (GP), the active form (GPa), and the inactive form (GPb). GP catalyzes the phosphorolysis of glycogen to release glucose‑1‑phosphate, which, through a coupled reaction involving PGM and G6PDH, reduces NADP⁺ to NADPH. The NADPH then reacts with a chromogenic agent to form a colored product with a characteristic absorption peak at 450 nm; the rate of product formation is directly proportional to the enzyme activity. The total GP activity is measured in the presence of 5′‑AMP, while the GPa activity is measured in its absence; the difference between the two values corresponds to the GPb activity.
