Lysine is one of the essential amino acids for humans. It promotes growth and development, enhances immune function, and plays a role in improving the function of the central nervous system. Because the lysine content in cereal foods is very low and is readily destroyed during processing, it is often referred to as the first limiting amino acid. The assay principle of this kit is as follows: lysine in proteins contains a free ε‑NH₂ group, which reacts with ninhydrin reagent to form a blue‑purple colored product. The color intensity is linearly related to the lysine content within a certain range. Since leucine has the same number of carbon atoms as lysine and contains only one free amino group (ε‑NH₂), leucine is commonly used to prepare the standard solution. The lysine content in the sample can be quantitatively calculated by measuring the absorbance.
