Overview
Methylmalonyl-CoA synthase is widely present in the mitochondrial matrix of animals, plants, microorganisms, and cultured cells. It works in conjunction with citrate synthase (CS) to participate in the regulation of the tricarboxylic acid (TCA) cycle, primarily involved in the metabolism of propionate.
Assay Principle: MCS catalyzes the condensation of propionyl-CoA and oxaloacetate to form methylmalonyl-CoA. The methylmalonyl-CoA is subsequently hydrolyzed to yield methylcitrate. This series of reactions drives the conversion of the colorless compound DTNB into the yellow-colored product TNB, which exhibits a characteristic absorbance at 412 nm.
